利用共免疫沉澱及質譜技術分析與毛果楊微管相關蛋白 SP1L1_8 之交互作用蛋白

被子植物受到重力的作用,會造成枝幹的彎曲,形成引張材。引張材會出現纖維素含量與纖維微管角度排列的改變,是研究纖維素生合成與微纖維傾斜角排列的適當材料。針對毛果楊 (Populus trichocarpa) 正常材與引張材的轉錄體分析,發現部分基因之轉錄異構體具有差異表達的現象,但合成纖維素之纖維素生成酶並沒有表達上的差異,而是有其他因子的調控。其中調控微管之微管相關蛋白有出現差異表達的現象,可以對應纖維素微纖維排列角度的改變,包括有微管相關蛋白SP1L1_8。因此,為了了解 SP1L1_8 蛋白與微管或其他微管相關蛋白的作用,與如何調控微管以影響引張材纖維微角的排列方向,將利用客製化

Anti-SP1L1_8 抗體 A/B,分析毛果楊中與 SP1L1_8 具有交互作用的蛋白。藉由基因選殖,成功表達 6 個 SP1L1 蛋白,而兩支 Anti-SP1L1_8 抗體經過西方墨點測試專一性,結果顯示兩支抗體只會與 SP1L1_8 蛋白反應,具有很高專一性。選擇 Anti- SP1L1_8 antibody 偵測毛果楊各個組織的粗蛋白,無法偵測到 SP1L1_8 蛋白,之後的共免疫沉澱以西方墨點測試與蛋白鑑定也無法偵測到該蛋白。因此修改實驗方法,在毛果楊粗蛋白萃取液中添加 SP1L1_8 蛋白後進行共免疫沉澱,以西方墨點偵測可以偵測到該蛋白,

但是質譜鑑定蛋白結果不理想,因此無法與實驗修改前之質譜結果比較,及鑑定交互作用蛋白。雖然目前關於 SP1L1_8 蛋白研究相對較少,但本研究提供一個良好的實驗方法來了解與 SP1L1_8 蛋白有交互作用的蛋白。

在電子競技項目中,公平性是一項非常重要的要素,若在競賽中使用作弊的手段取得勝利,這不單單是破壞了遊戲的公平性,更將是對電子競技的發展造成不小的影響。近年來隨著電腦硬體及人工智慧演算法的進步,在射擊遊戲中,出現了一種基於電腦視覺於人工智慧演算法應用的新型態作弊手段,相較於傳統侵入式的作弊手段更加難以偵測及防堵。本研究將整合使用MoveNet姿態檢測演算法,及YOLOv4-tiny物件辨識演算法,實作出一款基於電腦視覺於人工智慧演算法應用的輔助瞄準系統,並驗證及評估其成效,確認其對電子競技遊戲是否真實存在威脅,若確實存在威脅,希望藉以本研究讓遊戲開發商找出反制的手段,降低對遊戲產業造成的損害。
